Abstract

Sodium nitrite is food preservatif in processed meat and is usually often consumed by strikethrough. Excessive consumption of sodium nitrite can cause liver cell damage. Jeringau (Acorus calamus L.) is a herbal plant that contains flavonoids. The activity of flavonoids is hypothesized can be used as anti-inflammatory, analgesic and antioxidant agent. This research aimed to determine the effect of sodium nitrite (NaNO2) and jeringau rhizome (Acorus calamus L.) extract on morphology and histopathological changes of liver in rat (Rattus norvegicus L.). This study used a complete randomized design (CRD) consisting of 5 treatments (negative control: CMC Na 0.5%, positive control: sodium nitrite 50 mg/kg BW and CMC Na 0.5%, jeringau rhizome extract 250 mg/kg BW dose, 500 mg/kg BW dose, and 750 mg/kg BW dose) for 40 days. The examination of hydropic degeneration and necrosis used scoring method with Manja Roegnik Histopathology model. Histomorphometry on hepatic cell damage was analyzed using one-way ANOVA and Duncan's test at a significant level of 0.05 on SPSS 26. The results showed that the dose of 250 mg/kg BW and 500 mg/kg BW of jeringau rhizome extract was able to significantly improve hepatic histopathology damage (p≤0.05) compared to the control group. Jeringau rhizome extract could reduce the level of damage to hydropic degeneration and necrosis in the liver due to exposure to sodium nitrite.
